Materials and methods
Respective positive, negative, and experimental groupsolutions were prepared. The positive control group, with a pH of4, was prepared by mixing 10 drops of 1.00 % ATP, 10 drops of 0.20M KCl, 10 drops of 4 mM MgCl2, and 10 drops of deionized water. Anegative control group, with a pH of 5, was prepared by mixing 10drops of 0.20 M KCl, 10 drops of 4 mM MgCl2, and 20 drops ofdeionized H2O. Each solution was prepared ina clean, unlabeled bottle to reduce the bias. Pipettes were placedin each bottle. A table in a word document was made containing threecolumns, which included the positive, negative, and experimentalgroups to record the length of the fibers after applying thecontrol groups. Six rows were inserted to apply each control groupsix times.
A long segment of rabbit psoas muscle fibers was placedin a Petri dish of 50% glycerol. Fibers were placed on a cleanglass slide and were detached into a thin segment the width of ahuman hair. This segment was measured with a ruler placed under theglass slide and cut to 20 mm in length. The muscle segment waspositioned on a different cleaned, dry glass slide. The fibers werestraightened without stretching them. The segment was covered with3 drops of positive control solution away from the blindexperimenter. After 1 minute, the fibers were straightened gently,and the contracted length was measured by a blind experimenter, whodidn’t know what experimental group was used, using a ruler placedunder the slide, and the number of mm lined up was counted. Themeasurement was recorded in the table under the positive controlcolumn by someone who knew which control group was used.
