GNMENTS e Restriction Digest/Ligation worksheet Preview 0 2. In this set of labs, you will be restriction digesting lambda phage DNA and inserting that DNA into pUC19, the plasmid vector. Although lambda phage DNA has many restriction sites, only the sites for EcoRI (Figure 1) are mapped below (distance between the EcoRI sites noted in red). Note that four recombinant plasmids can be produced. Each recombinant plasmid will have a puC19 vector and one of the four possible inserts produced from the lambda phage digest. Using that information, draw out all possible recombinant plasmids that could be produced by the digestion and subsequent ligation.
