Background C-type lysozymes catalyze the hydrolysis of the 1, 4-beta-linkages between N-acetyl- D-glucosamine and N-acetylmuramic acid in peptidoglycan heteropolymers of the prokaryotes cell walls and between N-acetyl-D-glucosamine residues in chitodextrins. C-type lysozymes are closely related to a-lactalbumin in sequence and structure making them part of the same family. The sequence comparison of a-lactalbumin shows a strong similarity to that of lysozymes, specifically the Ca-binding c- lysozyme. However, the c-Lysozyme in this experiment does not bind Ca suggesting that the expected evolutionary history is that gene duplication of the c- lysozyme was followed by mutation. A search for c type lysozyme proteins in mammalian systems revealed Sperm lysozyme like protein (SLLPI) in mouse and human (HSLLP1). These proteins are implicated in the fusion of the sperm and ova. However, SLLP1 proteins do not exhibit the catalytic activities exhibited by the c- type lysozymes. The functional difference between the SLLP1 proteins and HEW lysozyme C are attributed to the mutations of catalytic residue glutamine at position 35 to threonine (Q35T) and residue aspartic acid at position 52 to asparagine (D52N).
