20. You are studying the possible interaction of two proteins, jaz and bop in response to stimulation with the growth factor IGF. Figure A shows Western blotting with anti-jaz and anti-bop antibodies in an experiment where you isolate the cytoplasm (C) and nucleus (N) of untreated cells (-IGF), and Figure B shows the same experiment after treating cells with IGF (+IGF). Next, you treat cells with IGF, followed by a chemical crosslinker which crosslinks proteins that are in contact with each other together. You conduct an immunoprecipitation assay (IP) of IGF stimulated cells. Results are shown in Figure C. You repeat this immunoprecipitation experiment without the chemical crosslinker, results of that experiment are shown in Figure D. Finally, Figure E shows results of probing the samples used in Figure D with the labeled antibodies. Your data support which of the following conclusions? A. Jaz and bop do not interact with each other in either the cytoplasm or nucleus. B. IGF treatment prevents interaction of jaz and bop in the nucleus but allows it in the cytoplasm A C Anti-bop Anti-jaz C N C N 00 I I -IGF I Anti-bop IP Anti-jaz IP Anti-bop WB Anti-jaz WB Anti-jaz Anti-bop с N C N OO T I +IGF C. IGF treatment causes indirect interaction of jaz and bop in the cytoplasm but not nucleus D. IGF treatment causes indirect interaction of jaz and bop in the nucleus E.
