Questions For "Challenge Questions" the purpose is to compose thoughtful and complete answers each based on your study of the course materials. It is most important for you to provide a sound reason for your answer than it is to be correct. There are often multiple correct lines of reasoning. As long as your response is supported by a sound argument based on learning of the concepts provided in this course, you will be in good shape. 1. What is a colony forming unit (CFU)? Define the term using your text. 2. An important aspect of any science is data interpretation and hypothesis generation. Examine the isopleth graphs below that show two different ways to enumerate bacteria gathered from environmental samples. These data are from a study done to look at annual microbial abundance and diversity in lake Washington. a. What do you see? In 1 or 2 paragraphs, describe the trends in each graph and make sure to compare and contrast the data. b. What biology could explain the discrepancies in these data? Describe 2 testable hypotheses that could explain the discrepancies in these data. Remember that the water samples to generate each figure are exactly the same. Assume that there were no experimental mistakes made in generating each measurement. c. Make sure give constructively critique of two of your classmates' hypothesis, and or revise your own in responses to their posts.

Water samples were collected and cells in that sample were stained with a non-specific stain then visualized under light microscopy and counted using a hemacytometer. Cell concentration is indicated by the key on the right.

The same water samples as in figure 1 were plated to CPS agar which was determined to be the best medium for culturing lake microbes in this study. Cell concentration is indicated by the key on the right. 3 x 10³ CFU/ml
