Practice Exercise:
1. A drug with an elimination half-life of 1 hour was given to a
male patient (80 kg) by IV infusion at a rate of 300 mg/hr. At 7
hours after infusion, the plasma drug concentration was
11μg/mL.
a. Calculate the VD.
b. Predict what body compartment this drug might occupy and explain
why you made this prediction.
2. Mr. Jones is admitted to the hospital with pneumoniae due to
gram-negative bacteria. The antibiotic tobramycin is ordered. The
VD of tobramycin in Mr. Jones is 40 L and the k is 0.8 hr-
respectively.
a. What maintenance dose should be administered IV every 6 hours to
eventually obtain average steady-state plasma concentrations of 4
mg/L?
b. If you wish to give Mr. Jones an IV loading dose to achieve the
therapeutic plasma concentration of 4 mg/L rapidly, how much should
be given?
