Week & Serial Dilation BIOL 1107K Serial Dilution of Food Dye Learning Objective: Perform serial dilutions to prepare solutions of specified concentrations from a stock solution a series, Often in the lab you have a concentrated solution but need it at a lower concentration. You can dilute the solution to obtain the concentration you want. But how? Serial dilation is common technique used to dilute a concentrated solution by diluting that is making a dilution of a dilution several times in a row. This technique is useful if you want to study a range of concentrations of a substance or if you want to prepare a 1:1,000,000 dilution. (You wouldn't add 1 mi to 999,999 ml) In this exercise, you will systematically dilute solutions of food dye with water. You will observe how the color intensity, or saturation, of each subsequent solution changes as it becomes more dilute. You will calculate the concentration of each of the diluted solutions Vocabulary concentration: The amount of a substance (solute) dissolved in a liquid (solvent) in a chemical solution. Units are usually in 16 (weight'volume) or molarity (moles/liter). dilution: The process of reducing the concentration of the sample. stock solution: Original sample solution of greatest concentration that needs to be diluted. (It is not always necessary to know the concentration of the stock) dilution factor : volume of sample transferred: total volume. (Total volume-volume of sample + volume of solvent) Examples (See attached PowerPoint for figures of example) a) 1:10 dilution factor-1/10-1ml sample (1ml sample +9 ml solvent) b) 1:2 dilution factor=% 1ml sample (Iml sample + 1 ml solvent) NOTE: There is no 1:1 dilution Mixing equal parts is a 1:2 dilution Practice dilution factors: ++ 1 mL 2mg/ml BSA+4 ml water.
