Questions. 1. The concentration of alginate used to immobilize the enzyme was 3% (w/v). What do you think would be the effect of significantly increasing the concentration of alginate used? 2. What would be the likely effect of significantly speeding up the flow rate of the sucrose solution through the column? 3. How do you think you could experimentally determine the optimum flow rate through the column? 4. The substrate (i.e. 3% sucrose is made up in buffer at pH 4.7. If the sucrose was made up in water at pH 7 the rate of catalytic conversion would be slower. Why?
