Why does this metabolic intermediate accumulate? Oxidative decarboxylation of malonate forms the metabolic intermediate. Malonate blocks substrate-level phosphorylation, leading to an accumulation of the substrate. Malonate activates the enzyme that forms the intermediate, leading to its accumulation. Competitive inhibition of an enzyme by malonate causes the intermediate to accumulate. Why does adding malonate to isolated mitochondria stop oxygen consumption?
