I conducted this lab and have attached the results. I tested sixknown amino acids and one unknown compound containing a mixture of2 amino acids. The eluent I used was three parts Isopropyl Alcoholand 1 part Ammonia. After conducting the chromatography, I sprayedthe TLC plates with Ninhydrin and added heat from a blow dryer tomake the spots appear. The Spots on the TLC plates are thesame as the table going from left to right. 1. Alanine 2.Isoleucine 3. Lysine 4. Proline 5. tryptophane 6. Valine 7.Unknown. 8. Unknown.
I determined the Unknown compound to be a mixture ofAlanine/Valine. I have a few questions I need help answering todiscuss them intelligently.
1.Why are the RF values for the results what they are, and doesthat match what they should be for the theoretical structures ofthe Amino acids?
2.What is the difference in the polarity of the amino acids toone another and the eluent?
3.Why are the spots the color they are? in correspondence to theamino acids “order,” … i.e., primary amine, secondary amine,etc
