Capsule and Endospore Stain (3-8 and 3-9; 4 points) Purpose: You completed capsule stains and endospore stains and viewed the bacterial cells using the oil immersion lens (1000X magnification) in order to determine the presence or absence of certain cellular structures in a variety of bacteria. These stains can be used in identification of the organism and they provide information about the potential pathogenicity of a given strain of bacteria. The endospore stain reveals structures made by some bacteria and not others. It can yield important clinical information and informs us about the possible type of environment in which the organism may be found and the lengths to which disinfection of said environment may need to occur. Task: Read section 3-8 (pages 201-202) and 3-9 (pages 205-208) of your lab manual. You performed the capsule stain and the endospore stain on "model" bacteria in order to assess their cell morphology. . Using your lab manual, analyze and interpret your stains. ● Answer the following questions in your own words. • Upload this Word or PDF document containing your bolded answers directly into Canvas Criteria for Success: . .
1. Embed your photographs of the capsule stain in the space below (not at the end of the document, not uploaded separately). Label your photos well. For every photo, include the name of the bacterium, the type of stain you used, total magnification, and "photo by (insert your name)." Photos of Capsule Stains (3-8): . S. aureus B. megaterium B. subtilis K. pneumoniae
Using your stains and your photos from the lab, complete the following data table (if you did not perform a specific stain, indicate "N/A". Adjust the bacterial names if necessary to reflect what we used in lab): Organism (full Latin name, in italics) S. aureus B. megaterium B. subtilis K. pneumoniae Cellular Morphology and Arrangement (include a detailed sketch* of a few representative cells) Approximate Cell Dimensions, length x width (including the appropriate units) Approximate thickness of capsule (including the appropriate units) "Describe & sketch what YOU see. I want to know your interpretation. Capsule (+/-)
2. Embed your photographs of the endospore stain in the space below (not at the end of the document, not uploaded separately). . Label your photos well. For every photo, include the name of the bacterium, the type of stain you used, total magnification, and "photo by (insert your name)." Photos of Endospore Stains (3-9): B. subtilis (5 days) B. megaterium (48 hrs)
