The structure of simple polysaccharides can be determined by acombination of permethylation, hydrolysis, reduction, and in somecase enzymatic digestion. During permethylation (also known asexhaustive methylation) all the free hydroxyl residues aremethylated. Upon acid hydrolysis, the glycosidic bonds and themethyl groups attached to anomeric carbons are cleaved while theother methyl groups resist hydrolysis.
A trisaccharide of unknown structure is submitted topermethylation followed by acid hydrolysis. After this treatment,you identify three molecules:
2,3,4,6 tetramethylglucopyrannose
2,3,4 trimethylglucopyrannose
2,3,6 trimethylgalactopyrannose
The numbers indicate the methylated carbons in each of thesemolecules. Additional experiments show the following:
- The trisaccharide is reduced by the Fehling’s solution.
- The digestion of the trisaccharide by a class of enzymescalled β-glucosidases result in the complete digestion of thetrisaccharide and the release of two molecules of glucose and onemolecule of galactose. A βglucosidase cleaves a glycosidic bondthat is located downstream (on the right) of a glucose unit it aβconfiguration.
