Objectives: Upon completion of this assignment students should be able to.... 1. Describe the differences between the T and R-state of hemoglobin. 2. Predict the differences in hemoglobin oxygen-binding behavior of both native and mutated forms and portray these differences graphically. 3. Explain the relationship between protein cooperativity and conformational changes. Instructions This assignment is meant to be completed AFTER you have done the "Hemoglobin Conformational Change" structural tutorial and accompanying quiz AND while you are walking through "BC351- LN08D-Hb Worksheet" in Echo360 on Canvas. In the first minutes of this echo360 recording I demonstrate to the class a "popsicle model" of Hb. Once you have listened to this please proceed to activity #1 of this worksheet. Activity #1 Study the model and describe below what each of the listed parts (or actions) of the model represents on an actual hemoglobin model. In other words, what parts represent the subunits or the heme, etc. After you have finished with this consider the last questions. (HINT: This will be difficult to complete if you haven't completed the "Hemoglobin Conformation Change" in the "Structural Tutorials" tab of the book). After completing activity one listen to LN08D starting at-25:00. 1. Each popsicle stick: 2. The rubber band when not being pulled on: 3. The rubber band when being pulled on: 4. The paper clip: 5. The contact between the paper clip and stick #2: 6. The model as a square: 7. The "collapsed" model: 8. Your fingers pulling on the rubber band: Will this model be "cooperative"? Why or why not?
Based on your answer to the above question illustrate oxygen binding behavior of this model on the graph below.. % saturation po₂ Now that you have completed Activity #1 go the 25-minute mark in the echo360 recording (LNOSD) and listen while I explain the "take away" from this activity and then proceed to Activity #2. Activity #2 To start this activity begin listening to the echo360 recording at the -36-minute mark. What you are being asked to do is to attempt to form the "square" between the popsicle sticks again with this "mutated" (bent) paper clip (image below). Once you have listened to these resources answer the following questions based on this exercise. 1. What state (R or T) does this mutated model "favor"? Why? In explaining make sure you address what the mutation has changed from list of 8 things in activity #1.
2. Will this model be "cooperative"? Why or why not? 3. Based on your answer to the above question illustrate oxygen binding behavior of this (the mutated) model and the original model on the graph below. % saturation p0₂ Now that you have completed Activity #2 go the 44:50 mark in the echo360 recording (LN08D) and listen to the rest of the recording while I explain the "take away" from this activity. After doing so please submit your completed worksheet to the "Chapter 8: Hb Structural Tutorial Worksheet" assignment on Canvas. To do so you can simply print this off, write in your answers, and then scan it and upload it. Either that or use a pdf annotation software on a tablet. Either submission will work.
