Gas chromatography (GC) is a separation technique that often has very high chromatographic resolution and low bandwidth compared to liquid chromatography (LC). Nevertheless, there are major limitations with GC as a separation technique, not least in pharmaceutical analytical chemistry. Briefly write down three important limitations to the use of GC as a separation technique.I have the answer down below, but I dont understand it, can someone explain in easier way?Answer:Gas chromatography can only be used for compounds that are relatively volatile and non-polar. Most drug substances are polar and non-liquid.Derivatization reactions can be used toenable the separation of polar compounds with GC but this then requires an additional sample processing system.GC is also not suitable for analysis of water-based samples and solutions and most drug substances are best soluble in water.More generally, GC is considerably less flexible as a separation technology than LC because the mobile phase in GC is inert and one cannot effectively optimize the separation by varying the mobile phase.
