The Unknown Project is a capstone project where you will apply many of the skills you have learned this quarter to identify the bacterial organisms in a broth suspension. You will be expected to identify the organisms in your sample to Genus and species names. The organisms in the samples are organisms that you have worked with at some point in the quarter. Here is how we will do this lab project virtually: Part 1: (10 points) You will receive an unknown number with the first round of data via email. Review and interpret the data in your sample unknown. Record the data in the data sheet posted in Canvas. Part 2: (20 points) After reviewing your results from Part 1 consider how you would proceed to identify your organisms in your sample. What tests would you need to do to identify your organisms? Submit a summary of the tests you plan to do and how these tests will allow you to identify your organisms. You can reference any labs for ideas of biochemical tests, refer to the labs that we have completed which have similar organisms to your unknown sample. Submit your summary plan (be sure to include the specific biochemical tests you plan to do) to Canvas. Once you have submitted your plan you will receive the second round of data via email. This second round of data will consist of the biochemical tests you have requested. You should be able to identify your unknowns and determine the organism to the Genus and species name. Part 3: (20 points) Submit your results and include a summary of how you identified your organism. For each organism in your unknown sample include a brief description of the microorganism with the following information: where the organism is typically found, is it a pathogen or part of normal microbiota, what diseases are caused by the organism, identify virulence factors that promote disease, what treatments are available, and whether or not there are vaccines available.
Which organisms are possible choices for the Unknowns? Here is a list of possible organisms: Staphylococcus aureus Staphylococcus epidermidis Micrococcus luteus E. coli E. aerogenes Klebsiella pneumoniae Pseudomonas aeroginosa Citrobacter freundii Alcaligenes faecalis Salmonella enterica Shigella sonnei Proteus mirabilis
Day 1A. Prepare a gram stain of broth "Unknown Sample". Note: How many organisms do you see? What is the gram reaction, shape and arrangement of organisms? Day 1 B. Inoculate your "Unknown Sample" to TSA, MacConkey, and Mannitol Salt agar plates using streak technique. Incubate at 37oC for 24 hours. Consider what types of organisms you can expect to grow on these three different types of media. Are any selective, differential, minimal agars? Day 2.
