leukocyte - Selectins are a class of proteins that specifically interact with carbohydrates. They have a pattern of invariant and highly conserved amino acid residues forming the carbohydrate recognition domain (CRD). The selectin family of adhesion molecules mediates the initial attachment of leukocytes (white blood cells) to vascular endothelial cells at sites of tissue injury and inflammation. The image at right depicts a leukocyte interacting with a selectin embedded in the membrane of a vascular endothelial cell. a. What molecules are shown covalently bound to the surface of the leukocyte? selectin vascular endothelial cells b. Of these molecules, which most directly mediates the interaction of the leukocyte with the selectin? fructose N-Acetylglucasamine galactose N-Acetydneuraminic acid c. Draw the structure of this molecule. d. What type(s) of noncovalent interactions could be occurring between the molecule and the selectin? e. Therapeutic agents could block the action of selectins in order to prevent the pathological effects resulting from leukocyte entry into sites of inflammation. These agents would block selectin binding to target molecules. Propose a molecular structure for an effective therapeutic agent.
