Due to steric hindrance, the bond indicated by the red arrow has
high barrier to rotation and the equilibrium or the rotomers as
shown below does not put the nitrogen group into the correct
position for binding. What changes could you make to lock the
orientation of the compound to ensure that the binding sites line
up?
