Hello, here is the question I was asked:
"Designan experiment to determine whether a particular protein istransported anterogradely orretrogradely in axons. (1) State what experimentalmethod/strategy/technique you would use; (2)describe what manipulations you would make, if any, and what youwould measure; (3) include anexplanation of how you would interpret the resulting data (e.g.what result would indicate anterogrademovement and what result would indicate retrograde movement. (Youshould be able to do this with 1-2sentences per point above.)"
This is my answer so far:
"The technique that can be used to study the retrograde andanterograde transport can be studied using Radioactive tracers, andthe same can be identified using autoradiography. The size andextent of many neurons would be challenging to study the transportmechanism, so the neurons can be segmented, and those segments canbe radioactively labeled. Interpretation of data: the amount ofradioactivity can be identified by fluorography and the segmentscan be analysed by gel electrophoresis. The type of transport canbe identified"
These are the key points that in some way each needed to beaddressed:
"Key points:1) (6 points) Here are some possible approaches (a), withmeasurements (b) and interpretations (c). Youneed all three parts (a, b and c) for a complete answer.(a) Fluorescently tag the protein, then do imaging. (b) Measurewhether the protein movesaway from or toward the soma. (c) if the protein moves away fromthe soma, it's movinganterograde; toward the soma is retrograde.(a) Perform ligation of an axon. (b) Measure whether the proteinaccumulates on the proximalor distal side of the ligation. (c) if the protein accumulates onthe distal side (farther from thesoma), it was moving retrogradely; if the protein accumulates onthe proximal side (closer to theanal ima mauing anterogradely"
I already completed and turned in this assignment but I want tounderstand more about how I could improve my answer even though itwasn't supposed to be a super long answer (as this could all comeup again in class).
Please help me understand the key points I answered and the keypoints that you believe I need to look further into/missed.What could satisfy that point instead?
