Phenotype for each individual
You are studying the "University" trait in humans. This trait is governed by the branched metabolic pathway shown below. The proteins A, B and D act at steps in this pathway as indicated in the diagram. High school students who produce the molecule Z are very intelligent and attend UIUC (the Illinois phenotype). The intermediate Y inhibits brain function and students who do not make Z and accumulate Y end up at Yale (the Yale phenotype). The intermediate X inhibits brain function more severely, and students who don't make Y and accumulate X, end up at Stanford (the Stanford phenotype). The enzyme D converts X to W only in the absence of A (A is a more efficient enzyme than D). Students who make W are most impaired and end up at Indiana (the Indiana phenotype). There are only two alleles of each of these genes, one wild-type allele and one complete loss of function allele. The two alleles of each are distinguishable by a polymorphism that is detectable by ASO microarrays as shown in the figure below. You extract the DNA from eight individuals (labeled 1-8 on the figure below) and run an ASO microarray. Your microarray contains DNA complementary to the three genes A, B and D as indicated in the figure. The color scheme for the diagram of the arrays below, is the same as that discussed in lecture. In this diagram R indicates a red spot, P a pink spot, and B a black spot. Individual 1 has the Illinois phenotype and individual 2 has the Indiana phenotype.
