BACTERIA FROM THE ENVIRONMENT - CONSULT YOUR INSTRUCTOR BEFORE BEGINNING THIS SECTION TO SEE IF THERE IS TIME TO COMPLETE- In this exercise, we will sample various environments to observe the bacteria/fungi that are present. We will use tape to collect our sample and we will transfer this sample to an agar plate. Agar plates contain nutrients that will allow any bacteria or fungi present in our sample to multiply to the point where they become a visible colony. Each group (consisting of three or four students) will be given four agar plates. As a group, you will decide on four different environmental surfaces in which you will collect your samples from (i.e., elevator button, cell phone, lab desk, lab floor, door knob, toilet handle, soap dispenser, pop machine button, coin, etc.). To collect your sample, fold the end of a piece of cellophane tape over so that you have a non-sticky surface to hold. Place the sticky end on the environmental surface that you wish to sample. Pull the tape from the surface and press lightly on the agar surface of the plate. BE CAREFUL not to touch the agar surface with your finger, and keep the lid of your plate closed as much as possible to avoid contamination. With a felt pen, write your name on the BOTTOM of your agar plate, your section number, as well as the environmental surface sampled. DO NOT label the lids of the agar plate! The bottom half of the plate is where the agar is found. The agar plates will be incubated upside down at 37 °C overnight to allow microorganisms to multiply. Plates will be brought to your lab next class so you can make observations. Depending on the environment to which you exposed the plates, you may see an abundance of different bacterial and/or fungal species. Most bacterial colonies are typically white, cream or yellow in appearance and are relatively circular.
