The goal of this laboratory exercise is to explore enzyme specificity as well as the effects of several factors influencing enzyme activity (temperature, enzyme concentration, substrate concentration), Introduction Cell metabolism refers to the sum of all chemical reactions carried out by a living cell. Many of these chemical reactions require the presence of enzymes. Enzymes are proteins that function as biological catalysts, speeding up the rate of chemical reactions in living systems. Today you will be working with the enzyme catalase. Catalase speeds up the following reaction: 2 H₂O2 2 H₂O hydrogen peroxide water oxygen The enzyme is not listed as a product or a reactant, because it is not consumed or produced during the chemical reaction. Once the products have been formed, catalase is free to react with additional hydrogen peroxide molecules. Question 1 catalase Water 02 In a chemical reaction, the substrate is the substance upon which the enzyme acts. What is/are the substrate/s of catalase? Choose all that apply. Hydrogen peroxide Oxygen 1 pts
The products of the chemical reaction are results of a chemical reaction. What is/are the product/s of the chemical reaction above? Choose all that apply. Water Oxygen Hydrogen peroxide Here are the results of the first lab activity, dealing with how catalase normally functions. Answer the following questions based on the information provided in the table below: Data Table: Catalase Activity Tube 1 2 3 Contents Catalase Hydrogen peroxide Water Hydrogen peroxide Catalase Sucrose solution Bubble column height (mm) 7 0
D Question 3 Look at the tube containing catalase and hydrogen peroxide. Look at the equation for the chemical reaction again. Which product is responsible for the bubble column? O Hydrogen peroxide Catalase Oxygen O Water Question 4 1 pts Yes Look at the tube containing water and hydrogen peroxide. Will a chemical reaction occur when the enzyme, catalase, is not present? No 1 pts
Question 5 Look at the tube containing catalase and sucrose solution. Will the enzyme work with a substrate other than hydrogen peroxide? Yes No Here are the results of the second lab activity, dealing with how temperature affects catalase function. Answer the following questions based on the information provided in the table below: Effect of Temperature on Catalase Activity Tube Location 1 N 3 Cold water bath Room Bubble column height (mm) Temperature Boiling water bath 2 11 1 pts 0
D Question 6 Based on the results, what is the best temperature for catalase activity? Cold water Boiling water Room temperature Question 7 1 pts 1 pts Why there was a difference in the chemical reaction rate in the cold tube as compared to the room temperature tube? There was no difference between the chemical reaction rates in the two tubes. The molecule movement slowed down in the cold tube, decreasing the enzyme-substrate collisions, which decreased the rate of the reaction. The enzyme was denatured, which slowed down the rate of the reaction. The molecule movement increased in the cold tube, increasing the enzyme-substrate collisions, which increased the rate of the reaction.
Question 8 Why there was a difference in the chemical reaction rate in the boiled tube as compared to the room temperature tube? The molecule movement increased in the boiled tube, increasing the enzyme-substrate collisions, which increased the rate of the reaction. The molecule movement slowed down in the boiled tube, decreasing the enzyme-substrate collisions, which decreased the rate of the reaction. The molecule movement increased in the boiled tube so much that the reaction could not occur. O The enzyme was denatured, which slowed down the rate of the reaction. Hare the results of the second lab activity, dealing with how changing concentration of substrate and enzyme affects catalase function. Answer the following questions based on the information provided in the tables below: Substrate Concentration Enzyme Concentration Bubble Column Height (mm) X 1x 4x 1 x 1x 1x 8 12 1 pts 62
D Substrate Concentration 1x 1 x 1x Question 9 Enzyme Concentration [Select] ¼ X 1 x 4 x Bubble Column Height (mm) 2 enzyme concentration. 9 40 In theory, what concentration of substrate and what concentration of enzyme used in lab, when mixed, would give the fastest chemical reaction rate? (Hint: it may be a combination not done during the lab) The answer is [Select] substrate concentration and 1 pts
Question 10 In theory, what concentration of substrate and what concentration of enzyme used in lab, when mixed, would give the slowest chemical reaction rate? (Hint: it may be a combination not done during the lab) The answer is [Select] [Select] substrate concentration and 1 pts enzyme concentration.
