You are studying a family, all of whom are deficient in the synthesis of adenosine, requiring that all adenosine be derived from their diet. You sequence the DNA for the genes in the pathway - they are all normal. You probe for the mRNAs; all are present at normal levels except one, which is undetectable. The nucleosome modifications of acetylation and methylation indicate that these genes are all transcriptionally active, however. Previous work has shown that this undetectable mRNA is translationally controlled by a miRNA. What happened genetically in this family? (10 points) How can you prove it? (10 points)
