You are studying a family, all of whom are deficient in the synthesis of adenosine, requiring that all adenosine be derived from their diet. You sequence the DNA for the genes in the pathway - they are all normal. You probe for the mRNAs; all are present at normal levels except one, which is undetectable. The nucleosome modifications of acetylation and methylation indicate that these genes are all transcriptionally active, however.
