Using the data in Table 2, plot a single Lineweaver-Burk graph for the imaginase-catalysed reactions with no inhibitor (control) and in the presence of inhibitor A and then in the presence of inhibitor B; your graph should have all three plots (graph not to be included in your assignment). Then, use the plots to determine the K, and V.values for each set of data. Answer the following questions. (12 marks) a. Give the Vand K, values for the control (uninhibited) reaction and the reactions in the presence of each of the inhibitors. Don't forget the units b. Compare the K, and V_.values for inhibitor A to those of the controls. Were they the same or different? Explain any differences c. Compare the K, and V.values for inhibitor B to those of the controls. Were they the same or different? Explain any differences. 17. Respond to the following questions. (8 marks) 2. On the basis of your analysis and what you already know about the inhibitors of enzyme activity, decide whether inhibitor A is a competitive or a non- competitive inhibitor of imaginase. Explain your answer. 3. Using the same criteria as for part a decide whether inhibitor B is a competitive or non-competitive inhibitor of imaginase, and explain your answer.
