Please answer all parts of the question,
E. coli RNA polymerase holoenzyme is composed of core
enzyme and sigma factor. Dr. K identified a novel gene in E.
coli and named it as Cosmos. The DNA sequence below
is a part of Cosmos. It is the sequence of sense strand.
Dr. K determined that RNA polymerase containing sigma 70
transcribed this gene.
5’-GGATGCTTCAGCGGCGACCGTGATGCGGTGCGTCGTCAGGCTTGACAGTATGCATTGCA

Part A) Highlight the promoter region in the sequence above and
indicate the name of each element. Also identify +1 site.
Part B) Highlight the ribosome-binding site and explain why you
chose that region (1 point).
Part C) Multiple ATG codons are present in the given DNA
sequence. Highlight the most likely start codon and explain why you
chose that codon (1 point).
Part D) What is the predicted sequence of Cosmos protein?
Provide first 7 amino acids using one letter code given in the
codon table below (0.5 points).
Part E) Transcription and translation efficiency contribute to
the expression level of a gene. A mutation can increase the
expression of Cosmos. Which nucleotide(s) would you mutate to
increase Cosmos expression and why (1 point)?
