QUESTION 7 There's been a murder and four possible suspects are being questioned. A blood sample has been found under the fingernails of the victim, but the sample is a mixture of the victim's and the killer's. An SSR screen was run by genetic testing lab to identify which suspect is the killer. The testing lab said that the sequencing machine broke mid-analysis, thus the investigation team only has data from the first 3 SSRs. SSR1 can be either 204 or 188 base pairs long. SSR2 can be either 146 or 122 base pairs long. SSR3 can be either 102 or 88 base pairs long. In the table below, “- " means that the allele is present. Which suspect is not eliminated by these results: Sample Victim Chad Marion Murray Sophia Length 204 188 146 122 102 88 Chad Marion Murray Sophia All can be eliminated

QUESTION 8 You are interested in studying eukaryotic transcription factors and have developed an in vitro transcription system using a segment of DNA that includes a transcribed gene and its predicted upstream regulatory region. The transcription of this DNA occurs in a defined system in which you add purified RNA polymerase II and basal factors You perform a series of experiments that compare the efficiency of transcription in your defined system with the efficiency of transcription in a crude nuclear extract. You test the two systems with your WT DNA and various deletion mutants that you have generated (see figure below). The results of your study are shown in the table: DNA added Nuclear extract Defined system WT ++++ + -127 ++++ + -81 ++++ + -50 + + -11 Based on these data, in which region would you predict lies an enhancer? further upstream from -127 O further downstream from -11 -50 to -11 -127 to -81 -81 to -50

QUESTION 9 The genes described below are part of the yeast mating signal transduction pathway that signals the presence of extracellular alpha pheromone to the nucleus. These genes are listed in the order they function in the pathway.' STE3 encodes alpha pheromone receptor STE4 encodes the B subunit of a heterotrimeric G protein STE7 encodes a protein kinase STE12 encodes a transcription factor Loss-of function- mutations in any of these genes are sterile. In addition, gain-of-function mutations can be isolated in these genes. For which gene would a loss of function mutation suppress gain-of-function mutations in any of the other three genes. STEZ STE4 STE12 STE3
