A biochemical transformation is carried out in mixed batch
systems with enzymes immobilized on a flat carrier. The process is
carried out at 25oC. The size of the immobilized enzyme plates is
given as 2x2 cm. In addition, the mean linear flow velocity of the
solution on the plate is estimated as 1 m/s. In the experiments,
the Vm and Km values of the enzyme were calculated as 5x10-6
mol/s·mg enzyme and 1.9x10-3 mol/L, respectively. The substrate is
glucose and its oxidation is carried out with an immobilized
enzyme. The enzyme was immobilized on this surface at a rate of
1x10-4 mg/cm2. The initial substrate concentration in the batch
reactor is 8x10-3 mol/L. According to these data;
a) Calculate the diffusion coefficient of glucose in water at a
given temperature.
b) Calculate the mass transfer coefficient (kL) value for the
defined system.
c) Calculate the initial reaction rate for the defined
system by graphical method.
d) Calculate the initial reaction rate iteratively for the
defined system.
