Problem 4.1 (A, B) Fluorescence Microscopy Imagine that you are working in a BME laboratory and you have a collaborator who makes new cadmium-based quantum dot nanoparticles that could be used for imaging breast cancer. Your collaborator asks you to figure out whether these particles might be useful by performing cell line-based tests. She tells you that the nanoparticles have intrinsic far-red/Cy5 fluorescence (excitation/emission = 640 / 670 nm). In your lab, there is an epi-fluorescence inverted microscope (similar to the one as what used in the fluorescence microscopy Labster) equipped with the following four fluorescence filters and phase-contrast objectives. Assume the lab has all reagents (dyes, etc.) to measure subcellular localization and all necessary cell culture equipment and supplies. Filter names Excitation (max) Emission (max) 519 nm 570 nm FITC 495 nm Rhodamine 550 nm Cy5 649 nm 662 nm Hoechst 358 nm 460 nm A. Propose a cell line experiment to determine whether these nanoparticles can be useful for imaging human breast cancer-you will use fluorescent dyes and the fluorescence microscope. You will want to propose to use an appropriate type of cell (doesn't have to be a specific cell line name, just type), appropriate fluorescent dyes to see where the particles localize in cells (you can simply specify which organelles or cellular features they might bind to, but make sure you specify what fluorescent label they have!), and appropriate imaging conditions (filters for each dye, describe image processing and display). (8 points) B. Please also sketch an expected result, including control. You are welcome to use multiple colors with legend, or specify what area should be a specific color with labels. You are welcome to just sketch one cell for experimental and control images. (4.5 points)
